چکیده

The regulation of centrosome number is lost in many tumors and the presence of extra centrosomes correlates with chromosomal instability. Recent work now reveals how extra centrosomes cause chromosome mis-segregation in tumor cells.

Structure and evolution of the Cinful retrotransposon family of maize.

A maize cDNA clone was isolated by virtue of its intense hybridization to total maize genomic DNA, indicating homology to highly repetitive sequences. Genomic homologues were identified and subcloned from an adh1-bearing maize yeast artificial chromosome (YAC). Sequencing revealed that the expressed sequence was part of a Ty3-gypsy-type retrotransposon. We discovered and sequenced two complete ...

Cell death associated with abnormal mitosis observed by confocal imaging in live cancer cells.

Phenanthrene derivatives acting as potent PARP1 inhibitors prevented the bi-focal clustering of supernumerary centrosomes in multi-centrosomal human cancer cells in mitosis. The phenanthridine PJ-34 was the most potent molecule. Declustering of extra-centrosomes causes mitotic failure and cell death in multi-centrosomal cells. Most solid human cancers have high occurrence of extra-centrosomes. ...

Are centrosomes or aneuploidy the key to cancer?

A News of the Week article by Elizabeth Pennisi about centrosome replication (5 Feb., p. 770) points out that centrosomes "might be a key to cancer." Indeed, abnormal structures and numbers, that is, more than two, of centrosomes have already been found in nearly all cancers tested (1, 2). The idea that centrosomes might be a key to cancer originated with Theodor Boveri, who proposed almost a c...

Acentrosomal spindle organization renders cancer cells dependent on the kinesin HSET.

Centrosomes represent the major microtubule organizing centers (MTOCs) of animal somatic cells and orchestrate bipolar spindle assembly during mitotic cell division. In meiotic cells, the kinesin HSET compensates for the lack of centrosomes by focusing acentrosomal MTOCs into two spindle poles. By clustering multiple centrosomes into two spindle poles, HSET also mediates bipolar mitosis in canc...

Intercellular centrosome number is correlated with the copy number of chromosomes in bladder cancer.

Centrosome amplification, which may accelerate tumor progression through chromosomal instability, is frequently observed in human malignancies. The intercellular relation between the number of centrosomes and chromosomes, however, is poorly understood. Therefore, the relationship between centrosomes and chromosomal copy number in the same cells was investigated in bladder cancer. Centrosomes we...
